Spot method에 의한 Ames test에서는 test organism, S-9, molten agar와 미량의 histidine - bacteria로 하여금 mutation에 필요한 몇번의 cell division을 할 수 있도록 하기 위해 넣어주는 것이다 - 을 섞어 minimal agar plate에 붓고, test chemical로 적신 disc를 test plate의 가운데에 놓는다. 그래서 Ames test에서는 bacterial system이 mammalian test system에 더 근접하도록 하기 위해 activating enzyme의 source로서 liver homogenate S-9를 넣어준다.

Ames test,Salmonella Typhimurium ,Mutagen,Carcinogen,toxicological test 에 관한 내용입니다. Test organism은 histidine-negative(his-)인 Salmonella typhimurium의 영양요구체(auxotroph) 균주이다. Chemical의 mutagenic effect는 종종 organism의 enzymatic pathway에 의해 influence을 받아 nonmutagen이 mutagen으로 전환되거나 혹은 반대로 mutagen이 nonmutagen으로 전환되기도 하는데, 포유류에서는 이러한 toxification이나 detoxification이 간(liver)에서 일어난다. 이 균주는 back-mutation이 일어나 his+로 바뀌지 않는 한 histidine이 없는 배지에서 자라지 못할 것이다.레포트/기타
